Evaluation of allelic polymorphism of LEPR gene and its association with growth and carcass traits in Mazandaran native chicken

Introduction: This study aimed to study polymorphism of the exon 7 region of the LEPR gene and its association with growth and carcass traits in Mazandaran native chicken.
Materials & Methods: For this purpose, 200 male chicks of Mazandaran native fowls reared in the same conditions and all were slaughtered at 12 weeks of age, were used. Traits before and after slaughter were measured and recorded. Before the destruction of all birds, blood sample and extraction of DNA from samples were performed by Cinnagen kit. Then, the desired locus of gene was amplified using specific primers and genotyped using PCR-RFLP method by BsrI specific enzyme. Phenotypic and genotypic data analysis conducted using statistical software of SAS9.2.
Results: The frequency of each allele (+) and (-) were estimated to be 0.675 and 0.325 in LEPR locus, respectively. Hardy-Weinberg equilibrium study using Chi-square test showed that the study population in loci is not in balance. The results showed a significant association between LEPR gene genotypes and the traits of abdominal cavity fat weight, intramuscular fat and meat pH (P<0.05).
Conclusion: Based on the results of this research can be concluded that LEPR gene can be used as a candidate for growth and carcass traits in Mazandaran native fowls breeding programs.
