fa اثر پروبیوتیک باسیلوس سابتیلیس بر ایمنی زایی واکسن خوراکی دوگانه استرپتوکوکوس اینیه، استرپتوکوکوس آگالاکتیه ریزپوشانی شده با آلژینات/کیتوزان در ماهی تیلاپیای نیل (Oreochromis niloticus) Effect of Bacillus subtilis on immunogenicity of Streptococcus agalactiae and Streptococcus iniae bivalent oral vaccine encapsulated with Alginate/chitosan in Nile tilapia (Oreochromis niloticus) تخصصي Special پژوهشي Research <div style="text-align: justify;">مقدمه: استفاده از پربیوتیک ها به منظور افزایش ایمنی زایی و کارایی واکسن ها در آبزیان در دهه اخیر کاربرد زیادی یافته است. در این تحقیق اثر پروبیوتیک باسیلوس سابتیلیس بر ایمنی زایی واکسن خوراکی دوگانه استرپتوکوکوس اینیه، استرپتوکوکوس آگالاکتیه ریزپوشانی شده با آلژینات/کیتوزان در ماهی تیلاپیای نیل (Oreochromis niloticus) مورد سنجش قرار گرفت و  از نظر ایمنی زایی، کارایی اثرات آن بر شاخص های رشد و پارامترهای خونی مقایسه گردید. مواد و روش ها: ابتدا باکتری دوگانه استرپتوکوکوس اینیه، استرپتوکوکوس آگالاکتیه به روش Internal Emulsification با موفقیت ریزپوشانی گردید. سپس ۶۰۰ قطعه ماهی تیلاپیای نیل با میانگین ۲/32±۳۰ گرم به ۱۰ گروه (هر گروه در سه تکرار ۲۰ عددی) شامل :A گروه ایمن شده با S. iniae، B: ایمن شده با S. agalactiae، C: ایمن شده با واکسن دوگانه، D: ایمن شده با S. iniae و باسیلوس سابتیلیس، E: ایمن شده با S. agalactiae و باسیلوس سابتیلیس، F: ایمن شده با واکسن دوگانه و باسیلوس سابتیلیس، G: ایمن شده با S. iniae ریزپوشانی شده و باسیلوس سابتیلیس، H: ایمن شده با S. agalactiae ریزپوشانی شده و باسیلوس سابتیلیس، I: ایمن شده با واکسن دوگانه ریزپوشانی شده و باسیلوس سابتیلیس، Control: گروه شاهد، تقسیم گردیدند. ماهی ها به مدت دو ماه در شرایط مشابه، تغذیه و نگه داری شدند و در روزهای صفر، ۳۰ و ۶۰ ضمن زیست سنجی ماهی ها، نمونه از هر تیمار اخذ و خونگیری و جداسای سرم انجام شد. شاخص های رشد و برخی شاخص های ایمنی و خونی ماهی بین تیمارها مقایسه گردید. سپس ماهیان هر گروه با باکتری زنده استرپتوکوکوس اگالاکتیه و استرپتوکوکوس اینیه چالش داده شدند و تلفات تیمارها تا ۱۴روز ثبت و بین تیمارها مقایسه صورت گرفت. نتایج: نتایج نشان داد شاخص های رشد تحت تاثیر ایمن سازی قرار گرفته و شاخص های رشد شامل ضریب تبدیل غذایی، ضریب رشد ویژه و درصد افزایش وزن در تیمار ایمن شده با واکسن دوگانه ریزپوشانی شده و باسیلوس سابتیلیس، به طور معنی داری نسبت به تیمار شاهد  بهبود یافته بود (۰/05>p). در بین شاخص های ایمنی، عیار آنتی بادی ضد استرپتوکوکوس اگالاکتیه و ضد استرپتوکوکوس اینیه، میزان احیای NBT، میزان گلوبولین و پرتئین سرم و فعالیت لایزوزیم سرم به طور معنی داری در مراحل نمونه گیری در تیمار خوراکی ریزپوشانی و باسیلوس سابتیلیس بهبود یافته بود (۰/05>p). پارامترهای خونی شامل: هموگلوبین، هماتوکریت و گلبول های سفید خون بین تیمارهای تحقیق تفاوت معنی داری نشان دادند (۰/05>p). بررسی ها نشان داد که در هر دو مرحله  نمونه گیری روزهای ۳۰ و ۶۰، بیش ترین تعداد گلبول های سفید مربوط به تیمارهای ریزپوشانی شده و باسیلوس سابتیلیس بود (۰/05>p). کم ترین میزان تلفات در چالش با باکتری زنده استرپتوکوکوس آگالاکتیه و استرپتوکوکوس اینیه، به ترتیب با 36/7% و ۳۰% مربوط به تیمار ایمن شده با واکسن دوگانه ریزپوشانی شده و باسیلوس سابتیلیس بود که نسبت به تیمار شاهد (۱۰۰%) کاهش معنی داری را نشان داد (۰/05>p). بحث و نتیجه گیری: لذا می توان نتیجه گرفت که با توجه به نتایج مناسب ایمنی زایی پروبیوتیک باسیلوس سابتیلیس بر واکسن دوگانه ریزپوشانی شده در تحقیق جاری و مزیت های ویژه تجویز واکسن خوراکی، با تحقیقات تکمیلی امکان استفاده از این روش به عنوان یک جایگزین برای واکسن تزریقی وجود دارد.</div> <div style="text-align: justify;">Introduction: The use of probiotics in order to increase the immunogenicity and efficiency of vaccines in aquatic animals has been widely used in the last decade. In this research, the effect of Bacillus subtilis probiotic on the immunogenicity of the bivalent oral vaccine of Streptococcus iniae, Streptococcus agalactiae encapsulated with alginate/chitosan in the Nile tilapia fish (Oreochromis niloticus) was measured and in terms of immunogenicity, its effects on growth indicators and blood parameters were compared. Materials & methods: First, the bivalent bacterium Streptococcus agalactia and Streptococcus agalactiae were successfully encapsulated by the internal emulsification method. Then 600 pieces of Nile tilapia fish weighing 30±2.32 gr were divided into 10 groups (Each group in three repetitions of 20 numbers) include A: treatment group with S. iniae, B: treatment group with S. agalactiae, C: treatment group with bivalent vaccine, D: treatment group with S. iniae and Bacillus subtilis, E: treatment group with S. agalactiae and Bacillus subtilis, F: immunized with bivalent vaccine and Bacillus subtilis, G: immunized with encapsulated S. iniae and Bacillus subtilis, H: immunized with encapsulated S. agalactiae and Bacillus subtilis, I: treatment group with bivalent encapsulated vaccine and Bacillus subtilis, and Control. The fish were fed and kept under the same conditions for two months, and on days 0, 30 and 60, along with fish biometry, samples were taken from each treatment and blood and serum were isolated. Growth indices and some immune and blood indices of fish were compared between treatments. Then, the fish of each group were challenged with live bacteria Streptococcus agalactiae and Streptococcus iniae, and the deaths of the treatments were recorded up to 14 days and compared between the treatments. Results: The results showed that the growth indices were affected by immunization and the growth indices including food conversion coefficient, specific growth coefficient and percentage of weight gain in the treatment group with bivalent encapsulated vaccine and Bacillus subtilis were improved significantly compared to the control treatment (p>0.05). Among the Immune indicators, anti-streptococcus agalactiae and anti-streptococcus iniae antibody levels, NBT recovery rate, serum globulin and protein levels, and serum lysozyme activity were significantly improved in the sampling stages in the oral treatment of encapsulated and Bacillus subtilis (p>0.05). Blood parameters including: hemoglobin, hematocrit and white blood cells showed a significant difference between research treatments (p>0.05). Investigations showed that in both stages of sampling on the 30th and 60th days, the highest number of white blood cells was related to the encapsulated and Bacillus subtilis treatments (p>0.05). The lowest rate of deaths in the challenge with live bacteria S. agalactiae and S. iniae, with 36.7% and 30%, respectively, was related to the treatment group with bivalent encapsulated vaccine and Bacillus subtilis, compared to the control treatment (100%) showed a significant decrease (p>0.05).
