fa مطالعه تأثیر خوراک دهی با پروبیوتیک های لاکتوباسیلوس بولگاریکوس و لاکتوباسیلوس پلانتاروم بر روی سیستم ایمنی و محافظت در برابر پاتوژن آئروموناس هیدروفیلا در مدل ماهی زبرا Studying the effect of feeding Lactobacillus bulgaricus and Lactobacillus plantarum probiotics on the immune system and protection against the pathogen Aeromonas hydrophila in Zebrafish model تخصصي Special پژوهشي Research <div style="text-align: justify;">مقدمه: ماهی زبرا از نظر فیزیولوژی دستگاه گوارش، فارماکولوژیکی و ژنتیکی بسیار به پستانداران شبیه است. هم چنین شباهت قابل توجه سیستم ایمنی ماهی زبرا به پستانداران به عنوان یک مدل جایگزین برای مطالعه ایمنی ذاتی و ایمنی تطبیقی و هم چنین بدخیمی های ناشی از سلول های ایمنی می باشد. بنابراین این ماهی مدلی جذاب برای بررسی فرایندهای اساسی التهابی و آسیب به روده است که در این پژوهش از آن استفاده شده و نتایج به دست آمده، برای انسان قابل تعمیم می باشد. هدف از این مطالعه بررسی اثر خوراک دهی با دو سویه پروبیوتیبک لاکتوباسیلوس بولگاریکوس و لاکتوباسیلوس پلانتاروم به صورت جدا و در ترکیب با هم بر روی سیستم ایمنی، حضور باکتری ها  در روده و محافظت بر علیه پاتوژن آئروموناس هیدروفیلا در مدل ماهی زبرا که یک مدل موفق برای مطالعات بیولوژی رشد مهره داران، تحقیقات فیزیکی و زیست پزشکی، کلونیزاسیون باکتریایی روده و پاسخ های ایمنی معرفی شده است، انجام گرفت. مواد و روش ها: تست RT-PCR برای اثبات حضور باکتری های پروبیوتیک در روده و ارزیابی کمی بیان ژن های ایمنی انجام شد و بررسی مرگ و میر ماهی ها بعد از مواجهه با باکتری پاتوژن صورت گرفت. باکتری های پروبیوتیکی بعد از آماده سازی با غلظت  1۰۸ cfu/g به میزان 3 درصد وزن بدن ماهی ها به خوراک افزوده شدند و 2 بار در روز به 240 قطعه ماهی خورانده شد و بررسی آزمون ها به مدت 60 روز (روزهای ۰ ، 28، 56 و 60) انجام گرفت.  نتایج: بر اساس نتایج بالاترین بیان ژن باکتری لاکتوباسیلوس پلانتاروم در روده در تیمار LA.pba در روز 28 بود و بالاترین بیان ژن باکتری لاکتوباسیلوس بولگاریکوس در تیمار LA.ba در روز 60 بود. بالاترین میزان بیان ژن های ایمنی IL1β و TNFα به عنوان سیتوکین های التهابی در گروه نرمال بود و حتی پس از مواجهه با پاتوژن در روز 60، کم ترین میزان بیان این ژن ها در گروه LA.pba  بود. هم چنین بالاترین نرخ زنده مانی در گروه LA.pba دیده شد.  بحث و نتیجه گیری: نتایج به دست آمده نشان دهنده تاثیر مناسب استفاده از دو نوع پروبیوتیبک لاکتوباسیلوس پلانتاروم و لاکتوباسیلوس بولگاریکوس در ایجاد محافظت در برابر عامل بیماری زای آئروموناس هیدروفیلا است. هم چنین به نظر می رسد که استفاده از این دو پروبیوتیک در ترکیب با هم اثرات مفیدتری را در مدل ماهی زبرا به همراه دارد.   </div> <div style="text-align: justify;">Introduction: Zebrafish is very similar to mammals in terms of physiology of digestive system, pharmacological and genetics. Also, the remarkable similarity of zebrafish immune system to mammals is as an alternative model for studying innate immunity and adaptive immunity as well as malignancies caused by immune cells. Therefore, this fish is an attractive model for investigating the basic inflammatory processes and damage to the intestine, which was used in this research and the results obtained can be generalized to humans. The purpose of this study is to investigate the effect of feeding with two probiotic strains of Lactobacillus bulgaricus and Lactobacillus plantarum separately and in combination on the immune system, the presence of bacteria in the intestine and protection against the pathogen Aeromonas hydrophila in the zebrafish model, which is a successful model for studies on vertebrate developmental biology, physical and biomedical research, intestinal bacterial colonization and immune responses have been introduced. Materials & Methods: RT-PCR test was performed to prove the presence of probiotic bacteria in the intestine and quantitative evaluation of the expression of immune genes, and the mortality of fish after exposure to pathogenic bacteria was investigated. After preparation, probiotic bacteria were added to the feed at a concentration of 108 cfu/g at the rate of 3% of the body weight of the fish and were fed to 240 pieces of fish twice a day and the tests were evaluated for 60 (0, 28, 56 and 60) days. Results: According to the results, the highest gene expression of Lactobacillus plantarum bacteria in the intestine in LA.pba treatment was on day 28, and the highest gene expression of Lactobacillus bulgaricus bacteria in LA.ba treatment was on day 60. The highest level of expression of immune genes IL1β and TNFα as inflammatory cytokines was in the normal group and even after exposure to the pathogen on day 60, the lowest level of expression of these genes was in the LA.pba group. Also, the highest survival rate was seen in the LA.pba group. Conclusion: The obtained results indicate the appropriate effect of using two types of probiotics Lactobacillus plantarum and Lactobacillus bulgaricus in creating protection against the pathogen Aeromonas hydrophila.
