fa بررسی تأثیر گابا بر زیست‌توده و بیان برخی ژن‌های دخیل در مسیر بیوسنتز آستاگزانتین هماتوکوکوس پلوویالیس(Haematococcus pluvialis) تحت شرایط مختلف نوری Investigating the effect of GABA on the biomass and the expression of some genes involved in the astaxanthin biosynthesis pathway of Haematococcus pluvialis under different light conditions تخصصي Special پژوهشي Research <div> <table align="center" hspace="0" vspace="0"> <tbody> <tr> <td style="padding: 0in 12px; text-align: justify;" valign="top">مقدمه: هماتوکوکوس پلوویالیس، یک ریزجلبک سبز است که به دلیل توانایی آن در تولید مقادیر زیادی آستاگزانتین مورد علاقه صنعت است. از این ریزجلبک به‌عنوان یک سیستم تولید زیستی، برای تولید آستاگزانتین استفاده می‌شود. این پژوهش باهدف افزایش تولید آستاگزانتین در هماتوکوکوس پلوویالیس از طریق القاگر گابا در شرایط نوری مختلف انجام گردید. انتخاب یک القاگر و نور مناسب، این پتانسیل را دارد که بهره‌وری فتوسنتز را به حداکثر برساند و منجر به کاهش هزینه‌های کلی تولید شود. مواد و روش ها: سلول‌های ریزجلبک، بعد از عبور فاز رشد، در 6 تیمار متفاوت با سه تکرار برای گابا با غلظت‌های (صفر و 0/0001 گرم در لیتر) و زیر تابش نور LED با سه رنگ سفید، آبی و قرمز با چرخه خواب بیداری (16-8) تیمار داده شد.  پارامترهای میزان رشد سلول‌ها، کلروفیل، کاروتنویید، مقدار آستاگزانتین کل و بیان برخی از ژن‌های مسیر بیوسنتز آستاگزانتین (psy، lcy، bkt2) موردبررسی قرار گرفت. نتایج: نتایج نشان داد که نور آبی نسبت به نور سفید  بازده تقسیم و رشد سلولی را افزایش داد (2/64 برابر) و نور قرمز نسبت به نور سفید میزان بیوسنتز و انباشت آستاگزانتین را افزایش داد (4/94 میلی‌گرم بر گرم). هم چنین، گابا رشد سلولی، تولید آستاگزانتین را افزایش می دهد. با استفاده از گابا در نور قرمز، انباشت آستاگزانتین مقدار قابل‌توجهی بالاتر از نورهای سفید و آبی بود؛ ولی سرعت رشد و تقسیم سلولی کم تری نیز داشت. نتایج بیان ژن مسیر سنتز آستاگزانتی نیز نشان داد که گابا در نور آبی و قرمز، را به‌طور قابل‌توجهی القا می‌کند. بحث و نتیجه گیری: به طورکلی گابا با نور قرمز بیش ترین میزان تولید آستاگزانتین را ایجاد می کند بنایراین گابا به همراه نور قرمز می تواند به عنوان یک محرک رشد و تولید استاگزاتین استفاده گردد.</td> </tr> </tbody> </table> </div> <div style="text-align: justify;"></div> <div style="text-align: justify;">Introduction: Green microalgae contain a wide array of valuable metabolites that can potentially attract numerous biomanufacturing businesses. Among these microalgae, Haematococcus pluvialis, a unicellular freshwater microalga belonging to the Chlorophyceae class, is prevalent in numerous aquatic environments worldwide. It has gained recognition as the most abundant and auspicious reservoir for the industrial-scale synthesis of natural astaxanthin.  The effects of γ-aminobutyric acid (GABA) (0 and 0.0001 g/L) on Haematococcus pluvialis different light condition were investigated in order to find the best inducer and suitable light type (white, blue, and red light) for maximizing the photosynthesis efficiency and low-cost production. Materials & Methods: Microalgal cells, after growth phase, were subjected to the different treatments and cell growth rate, chlorophyll, carotenoids, total astaxanthin content, and the expression of some involved genes in astaxanthin biosynthesis pathways (psy, lcy, bkt2) were measured. Results: The results showed that the blue light increased cell division and growth efficiency (2.64-fold) and red light increased biosynthesis and accumulation of astaxanthin (94.4 mg/g). Additionally, using GABA enhanced the cell growth and astaxanthin production. GABA under red light resulted to a significantly higher accumulation of astaxanthin compared to white and blue light, although it had a lower cell growth rate and division. Gene expression results also indicated that GABA significantly induces psy and pds genes under blue and red light.
