fa مطالعه مورفومتریک و فیلوژنتیک جدایه های استرتاژیا (تلادورساژیا) در بزهای غرب و جنوب غرب ایران Morphometric and phylogenetic study of Ostertagia (Teladorsagia) isolates in goats from the west and southwest of Iran تخصصي Special مروری Review <div> <table align="center" hspace="0" vspace="0"> <tbody> <tr> <td style="padding: 0in 12px; text-align: justify;" valign="top">مقدمه: گونه های استرتاژیا (تلادورساژیا) با ایجاد تغییرات ساختاری و عملکردی در شیردان و کاهش تولید و مرگ و میر نشخوارکنندگان کوچک می تواند خسارات اقتصادی قابل توجهی به دامداری وارد کند. این مطالعه با هدف تعیین ویژگیهای ریختسنجی و فیلوژنی گونههای جدا شده از بزهای غرب و جنوب غرب ایران انجام شد. مواد و روش ها: طی سال 1401، در مجموع از 315 راس بز ذبح شده در کشتارگاه های غرب (استان آذربایجان غربی) (200 راس) و جنوب غرب (خوزستان) (115 راس) نمونه گیری شد. تعداد 320 نمونه استرتاژیا (210 نمونه از غرب) و (110 نمونه از جنوب غرب) از شیردان بزهای ذبح شده، با استفاده از استریومیکروسکوپ شناسایی و از سایر نماتدها جداسازی گردید. براساس پارامترهای مورفومتریک، تعداد 180 جدایه ازکرم های نر (140) و ماده (40) استرتاژیا (تلادورساژیا) که دارای شاخص های واضح بودند، آماده سازی و با روش های میکرومتری استاندارد تعیین گردید. در نهایت، 18 جدایه (12 نر و 6 ماده) از دو منطقه که دارای بیش ترین تفاوت فنوتیپی بودند با استفاده از ژن ITS2 و روش‌های مولکولی تعیین توالی شدند. نتایج: نتایج مورفولوژیکی و مولکولی نشان داد که جدایه های شناسایی شده شامل تلادورساژیا سیرکومسینکتا، تلادورساژیا ترفورکاتا و مارشالاژیا اکسیدنتالیس بودند. در مقایسه، برخی شاخص های کرم های نر بین سه گونه هر منطقه و نیز در بین دو منطقه اختلاف معنی داری وجود داشت (0/001>P). نسبت درصد مری به طول بدن تنها در گونه استرتاژیا اکسیدنتالیس در هر دو منطقه، تفاوت معنیداری وجود داشت (0/05P<). در کرم های تلادورساژیا (استرتاژیا) سیرکومسینکتا و استرتاژیا تریفورکاتا ماده اگرچه در برخی از شاخص ها اختلاف معنی داری وجود نداشت (0/05<P). اما در برخی از شاخص ها (طول پرده فرجی، فاصله پرده از ابتدای اویژکتور و فاصله پرده از انتهای اویژکتور) اختلاف معنی داری وجود داشت. بحث و نتیجه گیری: آنالیز فیلوژنتیکی و توالی های ITS2 حضور 4 ژنوتیپ تلادورساژیا سیرکومسینکتا، سه ژنوتیپ  تلادورساژیا تریفورکاتا و دو ژنوتیپ مارشالاژیا اکسیدنتالیس را نشان داد. نتایج این مطالعه ارتباط مستقیمی بین ژنوتیپ‌های جدایه‌های مختلف و ویژگی‌های تغییرات فنوتیپی آن ها نشان نداد. رابطه بین تغییرات فنوتیپی و ژنوتیپی نیازمند مطالعات تکمیلی بیش تری است. با توجه به آلودگی منطقه ای سه گونه شناسایی شده استرتاژیا، مدیریت برنامه کنترلی آن ها ضروری است.</td> </tr> </tbody> </table> </div> <div style="text-align: justify;"></div> <div style="text-align: justify;">Introduction: Ostertagia (Teladorsagia) species can cause significant economic losses to animal husbandry by causing structural and functional changes in the abomasum, reducing the production and mortality of small ruminants. This study aimed to determine the morphometric and phylogenetic characteristics of species isolated from goats in the west and southwest of Iran. Materials & methods: During 2023, a total of 315 slaughtered goats were sampled in Western (West Azerbaijan Province) (n:200) and Southwest (Khuzestan) (n:115) slaughterhouses. A total, of 320 Ostertagia (210 from the west) and (110 from the southwest) from the abomasum of slaughtered goats were identified and separated from other nematodes using a stereomicroscope. According to morphometric parameters, 180 Ostertagia (Teladorsagia) isolates of male (140) and female (40) worms, which had clear indicators, were prepared and determined by standard micrometric methods. Finally, 18 Ostertagia isolates (12 males and 6 females) from two regions that had the greatest phenotypic differences were sequenced using the ITS2 gene and molecular methods. Results: The morphological and molecular results showed that the isolates included Teladorsagia circumcincta, Teladorsagia trifurcata and Marshallagia occidentalis. In comparison, some characteristics of male worms were significantly different between the three species of each region and also between the two regions (P<0.001). There was a significant difference in the ratio of esophagus to body length only in Ostertagia occidentalis species in both regions (P<0.05). Although, there was no significant difference in Teladorsagia circumcincta and T. trifurcata female worms (P<0.05). But there was a significant difference in some indicators (the length of the vulva flap, the distance of the flap from the beginning of the ovijector, and the distance of it from the end of the ovijector). Conclusion: Phylogenetic analysis and ITS2 sequences showed the presence of four genotypes of Teladorsagia circumcincta, three genotypes of Teladorsagia trifurcata and two genotypes of Marshallagia occidentalis. The results of this study did not show a direct relationship between the genotypes of different isolates and the characteristics of their phenotypic changes. The relationship between phenotypic and genotypic changes requires more additional studies.
