The effect of plant flavonoids on memory disorders and oxidative stress determining in male Alzheimer's rats treated with streptozotocin

Introduction: Alzheimer's disease is one of the most common neurodegenerative diseases. Beta-amyloid plaques and neurofibrillary tangles are two important factors in Alzheimer's disease. Many cellular changes, such as oxidative stress, neuronal inflammation, and mitochondrial disorders are also seen in Alzheimer's disease, which lead to neuronal death. The flavonoids in the chamomile plant (Matricaria recutita) have great effects on disorders caused by brain diseases such as Alzheimer's disease due to the production of antioxidants.
Materials & Methods:  In this study, the effects of chamomile plant flavonoids on memory disorders in Alzheimer's rats were studied. For this purpose, the alcoholic extract of the collected chamomiles and chemical analysis of chamomile essential oil (GC/MS) were used to prepare and purify flavonoids. In this study, 56 adult male rats were divided into 7 groups including control - vehicle 1 (solvent of flavonoids) and vehicle 2 (solvent of streptozotocin drug) - Alzheimer's - flavonoid 20mg/kg - flavonoid 50mg/kg - flavonoid 100mg/kg - used. Diabetes was induced by injecting a single dose of 60 mg/kg streptozotocin intraperitoneally. Flavonoids were administered for 15 days. The shuttle box device was used to measure memory and learning and the delay time in the shuttle box was recorded.
The results were analyzed SPSS 22 software, ANOVA and Tokay tests. The significance of the data difference was considered at the p ≤ 0.05 levels.
Results: The results of the data analysis showed that doses of 50 and 100 mg/kg flavonoid extracts of chamomile plant caused significant changes in the duration of the delay in entering the dark section at p≤0.001 compared to the control group. And finally, it improves the avoidance memory in rats, which does this effect probably due to the presence of compounds such as quercetin and phytoestrogens. Also, oxidative stress parameters were significantly reduced in Alzheimer's groups treated with chamomile flavonoids. (P<0.001) Plant flavonoids are able to restore spatial memory function and oxidative stress parameters in groups treated with streptozotocin to normal levels.
Conclusion: Plant flavonoids can be used as an active biological substance to reduce memory and learning disorders in Alzheimer's samples, however, clinical studies are needed to prove the issue.
